- Abstract: sequence comparisons of previously identified chalcone synthase
(chs) cdna from various plant species showed that some parts of the
sequences have highly conserved. according to these conserved
sequences, several 20-25 bases in length oligonucleotides were
synthesized as primers. first strand cdnas were synthesized from
total rna of tobacco flower as template. a partial chs cdna of 866
bp segment was identified by polymerase chain reaction (pcr)
amplification followed by southern hybridization with 32p labeled
parsley chs cdna probe. the dna sequence of the pcr product showed
high homology with the chs genes of Antirrhinum majus, petunia
hybrida, petroselinum hortense, phaseolus vulgaris, arabidopsis
thaliana and sinapis alba l. the striking level of dna sequence
homology between tobacco chs cdna and other plants confirms that
the pcr product to be part of chs cdna.
